Sheen was born in Newport, Wales, to Irene, a secretary, and Meyrick Sheen, a personnel manager. He was raised in Wallasey and later Port Talbot, Glamorgan, alongside his younger sister. As a teenager, he was a keen footballer who was scouted for Arsenal's youth team, though he ultimately pursued acting. He participated in the West Glamorgan Youth Theatre and the National Youth Theatre of Wales before attending Neath Port Talbot College. Sheen moved to London in 1988 to train at the Royal Academy of Dramatic Art (RADA), graduating with a BA in Acting in 1991.
Throughout the 1990s, Sheen worked primarily in theatre. His early professional stage roles included When She Danced in 1991, Romeo and Juliet in 1992, Don't Fool with Love in 1993, Peer Gynt in 1994, The Seagull in 1995, and Henry V in 1997. He co-founded the Thin Language Theatre Company in 1991 and the production company The Foundry in 1998. Sheen earned Laurence Olivier Award nominations for his performances as Mozart in Amadeus (1998) and Jimmy Porter in Look Back in Anger (1999).
In the 2000s, Sheen expanded into screen acting and gained widespread acclaim for portraying biographical figures. He played British Prime Minister Sir Tony Blair in writer Peter Morgan's The Deal (2003), later reprising the role in The Queen (2006) and The Special Relationship (2010), earning BAFTA Award and Emmy nominations. His other biographical portrayals include comic actor Kenneth Williams in Fantabulosa! (2006), broadcaster David Frost in Frost/Nixon, and football manager Brian Clough in The Damned United (2009). He also received Olivier nominations for Caligula (2003) and Frost/Nixon (2006).
Starting in 2009, Sheen took on a wider variety of film and television roles. He appeared in high-profile fantasy and sci-fi films such as Underworld: Rise of the Lycans (2009), The Twilight Saga: New Moon (2009), Tron: Legacy (2010), Midnight in Paris (2011), and The Twilight Saga: Breaking Dawn – Part 2 (2012). On stage, he directed and starred in The Passion and played the title role in Hamlet at the Young Vic from late 2011 to early 2012. In 2013, he starred in the Showtime drama series Masters of Sex, earning a Golden Globe nomination.
Sheen's later television work includes playing an incarcerated serial killer surgeon in Prodigal Son (2019–2021), Aziraphale in the fantasy comedy series Good Omens (2019–2026), and Chris Tarrant in Quiz (2020). During the COVID-19 lockdown, he co-starred as himself in the quarantine comedy series Staged (2020–2022). In addition to his acting career, Sheen is recognized for his political and social activism, and he renounced his OBE in 2017.
