Her accolades include an Academy Award, three Golden Globe Awards, four BAFTA Awards, five Emmy Awards, a Tony Award, two Cannes Film Festival Best Actress awards, a Laurence Olivier Award, a Volpi Cup, the BAFTA Fellowship, an Honorary Golden Bear, a Screen Actors Guild Life Achievement Award, and the Cecil B. DeMille Award. In 2003, she was appointed a Dame Commander of the Order of the British Empire by Queen Elizabeth II.

Mirren was born in Hammersmith, London, to an English mother, Kathleen Rogers, and a Russian father, Vasily Petrovich Mironoff. Her mother's family included butchers who served Queen Victoria, while her father belonged to an exiled line of Russian nobility. Her paternal grandfather was an Imperial Russian Army colonel and diplomat stranded in London after the 1917 Russian Revolution. Her father, a viola player and civil servant, changed the family surname to Mirren in 1951. Raised in Leigh-on-Sea, Essex, Mirren attended Hamlet Court primary school, St Bernard's High School for Girls, and the New College of Speech and Drama in London.

At eighteen, Mirren joined the National Youth Theatre, playing Cleopatra in Antony and Cleopatra (1965), which launched her career. She subsequently joined the Royal Shakespeare Company (RSC), appearing in productions such as The Revenger's Tragedy, Troilus and Cressida, and As You Like It. She toured internationally with Peter Brook's International Centre for Theatre Research and played Lady Macbeth and Queen Margaret for the RSC. Her early stage work included West End roles in Teeth 'n' Smiles, The Seagull, and The Bed Before Yesterday, alongside an acclaimed performance in Measure for Measure at Riverside Studios in 1979.

Mirren made her credited film debut in Herostratus (1967) and had her first major film role in Age of Consent (1969). Her breakthrough screen role came opposite Bob Hoskins in The Long Good Friday (1980). She gained further film recognition in O Lucky Man! (1973), Caligula (1979), Excalibur (1981), The Mosquito Coast (1986), and The Cook, the Thief, His Wife & Her Lover (1989). Mirren won the Best Actress Award at the Cannes Film Festival for Cal (1984).

On television, Mirren gained widespread recognition playing Detective Chief Inspector Jane Tennison in ITV's Prime Suspect (1991–2006). The role earned her three consecutive British Academy Television Awards for Best Actress from 1992 to 1994, as well as two Primetime Emmy Awards. She also won Primetime Emmy Awards for her lead performances in the television films The Passion of Ayn Rand (1999) and Elizabeth I (2005).

Mirren earned Academy Award nominations for her film performances in The Madness of King George (1994), Gosford Park (2001), and The Last Station (2009), winning the Academy Award for Best Actress for her portrayal of Queen Elizabeth II in The Queen (2006). She reprised her role as Queen Elizabeth II on stage in Peter Morgan's The Audience (2013), earning a Laurence Olivier Award and a Tony Award. Her other Broadway Tony nominations include A Month in the Country (1995) and The Dance of Death (2002).

Mirren's subsequent film credits include The Tempest (2010), Hitchcock (2012), Eye in the Sky (2015), Trumbo (2015), the action film Red (2010) and its 2013 sequel, and four films in the Fast & Furious franchise. On television, her work includes roles in Door to Door (2002), Phil Spector (2013), Catherine the Great (2019), 1923 (2022), and MobLand (2025).
