Pinkett Smith was born in Baltimore, Maryland, to Adrienne Banfield-Norris and Robsol Pinkett Jr. Raised primarily by her mother and grandmother Marion Martin Banfield, she took early lessons in piano, tap dance, and ballet. She attended the Baltimore School for the Arts to study acting and dance, where she befriended classmate Tupac Shakur and friend Josh Charles. Following her graduation in 1989, she studied drama for a year at the North Carolina School of the Arts.

Pinkett Smith made her feature directorial and writing debut with The Human Contract in 2008. On television, she starred in and executive produced the medical drama Hawthorne (2009–2011) and portrayed Fish Mooney on Gotham (2014–2017). As a producer, she earned a Tony Award nomination for Best Musical for Broadway's Fela! in 2010, and she co-founded the media company Westbrook with her husband Will Smith in 2019.
Beginning in 2018, Pinkett Smith began co-hosting the Facebook Watch talk show Red Table Talk alongside her mother and her daughter, Willow Smith. For her work on the show, she won a Daytime Emmy Award, and Time named her one of the 100 most influential people in the world in 2021.
