He graduated from St. Thomas More School in 1974 and earned a Bachelor of Arts degree from Columbia University, where he served as the Class Day Speaker during commencement exercises in 2021.
Van Peebles made his screen debut in 1968 on the soap opera One Life to Live. In 1971, he appeared in the film Sweet Sweetback's Baadasssss Song, playing a childhood version of a character portrayed by his father, who also directed the project. After taking a break from acting through the remainder of the 1970s, he returned in the 1980s with roles in Exterminator 2 and The Cotton Club. He landed his first lead role in Rappin' (1985) and went on to appear in Heartbreak Ridge (1986) and Jaws: The Revenge (1987).
He made his television directorial debut with an episode of the 1988 series Sonny Spoon, in which he also starred, and later directed episodes of 21 Jump Street and Wiseguy. In 1991, he received a Directors Guild of America Award nomination for his direction of Malcolm Takes a Shot. That same year, he made his feature film directorial debut with the black gangster film New Jack City, in which he also co-starred. Produced on an estimated $8 million budget, the film grossed over $47 million domestically and became the highest-grossing independent film of 1991.
Throughout the 1990s and 2000s, Van Peebles actively directed and acted across multiple genres. He directed and starred in the Western Posse (1993) and helmed the 1995 film Panther, which won the Silver Leopard and Ecumenical Jury prize at the Locarno International Film Festival. As an actor, he appeared in films such as Highlander III: The Sorcerer, Solo, and Michael Mann's Ali (2001), earning an NAACP Image Award nomination for the latter. In 2003, he co-wrote, produced, directed, and starred in Baadasssss!, portraying his father during the creation of Sweet Sweetback's Baadasssss Song. The project earned him Black Reel Awards for Best Director and Best Screenplay.
Van Peebles also directed episodes for numerous television series, including Damages, Law & Order, Lost, Boss, Nashville, and Empire. He wrote and directed feature films such as the comedy We the Party (2012) and the action thriller Red Sky (2014), and directed the 2016 Second World War film USS Indianapolis: Men of Courage.
In 2017, Van Peebles created, produced, directed, and acted in the television series Superstition. His activism includes participating in a 2014 public service video supporting the District of Columbia statehood movement as part of the 51 Stars campaign. In 2026, he was honored with the Icon of Achievement Award at the Lexus Uptown Honors Hollywood Awards.
