Whitford was born on October 10, 1959, in Madison, Wisconsin, to Genevieve Louie and George Van Norman Whitford. He graduated from Madison East High School in 1977 before attending Wesleyan University, where he earned a Bachelor of Arts degree in English and theater in 1981. He subsequently studied drama at the Juilliard School in Manhattan as a member of Group 14.

His professional acting career began in the mid-1980s with screen appearances on television series such as The Equalizer and All My Children, alongside his feature film debut in 1986's Dead as a Doorman. Whitford made his Broadway debut in 1990 in Aaron Sorkin's play A Few Good Men, initiating a long-standing working relationship with the writer. During the late 1980s and 1990s, he built a versatile screen resume with supporting appearances in films like Adventures in Babysitting, Presumed Innocent, Awakenings, Scent of a Woman, Philadelphia, and Billy Madison.

Whitford achieved a major career breakthrough when he was cast as Josh Lyman in Sorkin's The West Wing in 1999. His performance during the show's seven-season run earned him a Primetime Emmy Award for Outstanding Supporting Actor in a Drama Series in 2001, and he also wrote two episodes for the series. Following the conclusion of the show in 2006, he continued to collaborate with Sorkin on Studio 60 on the Sunset Strip and starred in various television projects, including the comedy The Good Guys and guest roles on shows like The Mentalist.

In 2014, Whitford began a recurring role in the series Transparent, winning a Primetime Emmy Award for Outstanding Guest Actor in a Comedy Series in 2015 and garnering a fifth career Emmy nomination for portraying Magnus Hirschfeld in the same show. After joining the cast of the dystopian drama The Handmaid's Tale as Commander Joseph Lawrence, he won another Emmy for Outstanding Guest Actor in a Drama Series in 2019, becoming the first performer to win guest acting Emmys in both comedy and drama categories. He subsequently portrayed the character through 2025.

Alongside his television work, Whitford has appeared in feature films such as The Cabin in the Woods, Get Out, The Post, The Darkest Minds, and Godzilla: King of the Monsters. He has also remained active on stage, including a Broadway run in Boeing-Boeing and a 2023 stage production of Peter Pan Goes Wrong in Los Angeles.
