She was born in Montreal, Quebec, to Moroccan Jewish parents who observed Orthodox Judaism. Her mother was born in Casablanca, while her father originated from Rabat. When she was nearly two years old, her family relocated to Toronto, Ontario, and she was raised in the nearby suburb of Markham alongside her older brother and older sister.

Chriqui developed an interest in performing during her childhood after her brother funded her acting lessons. Her mother, who worked as an aesthetician, encouraged her creative pursuits before passing away when Chriqui was sixteen. She participated in the drama curriculum at Unionville High School, studied high school in Paris, and completed two years of comedy training at the Perimony school in France before committing to a professional acting career.

Her career began at age ten with an appearance in a McDonald's television commercial. In the mid-1990s, she moved to Vancouver, securing guest roles on shows such as Are You Afraid of the Dark?, Forever Knight, Once a Thief, and Psi Factor: Chronicles of the Paranormal. She made her Hollywood debut with a supporting role in Detroit Rock City in 1999, followed by a major role as Claire Bonner in the 2000 comedy Snow Day. She subsequently appeared in films including 100 Girls, Wrong Turn, On the Line, In the Mix, and National Lampoon's Adam & Eve.

In 2008, Chriqui portrayed Dalia alongside Adam Sandler in You Don't Mess with the Zohan and appeared as Revetta Chess in Cadillac Records with Beyoncé Knowles. Her other film projects include Women in Trouble, its sequel Elektra Luxx, and the 2019 release The Knight Before Christmas opposite Vanessa Hudgens. For her performance in 100 Girls, she was nominated for a DVD Exclusive Award, and she received a Teen Choice Award nomination for On the Line. In April 2008, she received the Standout Performance Trophy at the Young Hollywood Awards.

On television, Chriqui gained prominent recognition playing Sloan McQuewick on the HBO series Entourage, later reprising her role in the 2015 film adaptation. She also secured roles in Showtime's The Borgias, portrayed Lorelei Martins on CBS's The Mentalist, and played Lana Lang as well as her Bizarro counterpart Lana-Rho on the CW series Superman & Lois from 2021 to 2024. Her additional work includes voice roles as Cheetara in ThunderCats and Numbers in Call of Duty: Black Ops, as well as appearances in music videos for Hinder, Zac Brown Band, and Charles Perry.

Chriqui acquired U.S. citizenship in September 2017 and practices Transcendental Meditation. She has been in a relationship with actor Sam Trammell since 2020. Following the 2023 attacks on Israel, she spoke out against Hamas, signed an open letter with Creative Community for Peace calling for the release of hostages, and later signed a statement supporting Israel's participation in the 2024 Eurovision event.
