Shalhoub was born on October 9, 1953, in Green Bay, Wisconsin, where he grew up as the ninth of ten children in a Lebanese Christian family. His father immigrated to the United States from Lebanon as a child, later becoming a meat peddler, while his mother was a Lebanese American whose family ran a candy business. Introduced to performance when his sister entered him as an extra in a high school musical, Shalhoub pursued higher education at the University of Wisconsin–Green Bay and the University of Southern Maine, where he completed a bachelor's degree. He went on to receive a master's degree from the Yale School of Drama in 1980.

After graduating from Yale, Shalhoub spent four seasons performing with the American Repertory Theater before moving to New York City. He made his Broadway debut in a 1985 production of The Odd Couple and secured small film and television parts. His career breakthrough arrived in 1991 when he landed the role of Italian cab driver Antonio Scarpacci on the NBC sitcom Wings, remaining with the show until 1997. During this era, he also expanded into feature films, taking roles in titles such as Barton Fink, Big Night, Men in Black, Gattaca, The Siege, and Galaxy Quest.

In 2002, Shalhoub took on one of his most prominent television roles as the lead character in Monk. His performance earned him three Primetime Emmy Awards for Outstanding Lead Actor in a Comedy Series and a Golden Globe Award. Later, from 2017 to 2023, he portrayed math professor and activist Abe Weissman in the Amazon series The Marvelous Mrs. Maisel, winning a Primetime Emmy Award for supporting actor in 2019. He reprised his signature detective role in the 2023 television feature Mr. Monk's Last Case: A Monk Movie.

Alongside his screen career, Shalhoub has maintained a prominent presence in the theater. He earned Tony Award nominations for his performances in the stage plays Conversations with My Father, Golden Boy, and Act One. In 2018, he won the Tony Award for Best Actor in a Musical for playing Tewfiq Zakaria in the Broadway production of The Band's Visit. His stage repertoire also includes revivals of Lend Me a Tenor and The Price.

Shalhoub's extensive voiceover work includes playing the character Luigi in the animated movie franchise Cars, as well as voicing Splinter in two live-action Teenage Mutant Ninja Turtles films. In his personal life, Shalhoub married actress Brooke Adams in 1992, and the couple has two adopted daughters. Beyond acting, he co-founded the Arab-American Filmmaker Award Competition in 2005 and is an investor in a Michelin-starred restaurant.
