Raised in Miami, Florida, Murciano completed his secondary education at Christopher Columbus High School before attending Tulane University in New Orleans, Louisiana. He subsequently enrolled at the New England School of Law in Boston, but ultimately decided to abandon his legal studies to pursue a career in acting. Relocating to Los Angeles, he made his feature film debut in the 1997 action picture Speed 2: Cruise Control alongside Sandra Bullock.

In the early 2000s, Murciano secured roles in notable film and television productions, including the 2000 crime drama Traffic and the 2001 MTV soap opera Spyder Games, in which he played Francesco Torres. That same year, he appeared in the war thriller Black Hawk Down, where meeting producer Jerry Bruckheimer led to his breakthrough casting in Without a Trace. Portraying an agent in a New York City FBI missing persons unit, Murciano starred in the program throughout its seven-season run from September 2002 until May 2009. His performance earned him ALMA Award nominations in both 2008 and 2009.

Throughout his career, Murciano made frequent television appearances across various genres. He performed as Detective Carlos Moreno in the CBS police drama CSI: Crime Scene Investigation, initially appearing in a 2009 episode and returning for multiple episodes in 2012. Between April 2011 and January 2012, he portrayed CIA Agent Ray Cruz in the CBS series NCIS. Later in 2012, he took on the recurring role of Dr. Scott Evans in the ABC supernatural drama 666 Park Avenue. From 2014 to 2016, he appeared as Felipe Lobos in the Starz television series Power, and beginning in 2015, he joined the main cast of the Netflix series Bloodline as Detective Marco Diaz.

In feature films, Murciano reunited with Sandra Bullock in the comedy Miss Congeniality 2: Armed and Fabulous. His film work also encompasses roles in the dramas How to Go Out on a Date in Queens, The Lost City, and the Peruvian-Spanish production Máncora. He appeared in the 2014 science fiction feature Dawn of the Planet of the Apes as Kemp, and later played the gang leader Poison in the 2017 Netflix feature Bright. Additionally, he participated in music videos such as will.i.am's "Yes We Can" and the Demi Moore-directed video "Presidential Pledge."

Beyond his acting nominations, Murciano achieved public recognition in 2006 when People magazine named him seventh on its annual list of the Sexiest Men Alive.
