She holds the distinction of being the first female singer from Hong Kong to win the Golden Melody Award, an honor she has received three times. Over her career, Mok has released 18 solo studio albums, appeared in more than 50 feature films, and gathered over 15 million followers on the Chinese social media platform Weibo.

Raised in Hong Kong, Mok comes from a diverse ancestral background that includes Chinese, German, Persian, and Welsh heritage. Her grandfather was the first principal of King's College in Hong Kong. She attended Diocesan Girls' School, where she earned the 1st Hong Kong Outstanding Students Award, and later won a scholarship to the United World College of the Adriatic in Italy, graduating in 1989. She subsequently pursued studies in Italian Literature at Royal Holloway, University of London. Mok is fluent in English, Cantonese, Mandarin, Italian, German, and French.

While studying in London, Mok recorded music demos that earned her a contract with Star Records, leading her to return to Hong Kong and release her debut Cantonese album in 1993. She signed with Rock Records in 1996 under the mentorship of producer Jonathan Lee. She achieved her major musical breakthrough with her 1997 Mandarin album To Be, followed by her first major solo concert in Taipei in 2000. In 2003, Mok became the first female artist from Hong Kong to win the Golden Melody Award for Best Mandarin Female Singer.

Parallel to her singing career, Mok established herself as an actress. She gained widespread recognition for her performances in A Chinese Odyssey and Wong Kar-wai's Fallen Angels, earning Best Supporting Actress honors at the Hong Kong Film Awards, the Hong Kong Film Critics Society Awards, and the Golden Bauhinia Awards. Her international film appearances include Around the World in 80 Days, The Coffin, and Man of Tai Chi, alongside a return to Hong Kong cinema in the 2024 disaster film Cesium Fallout. She has also performed in musical theatre, playing Mimi in the Asian tour of Rent, and voiced animated dubs.

Mok expanded her music career through international tours, acting as creative director and producer. In 2013, she released Somewhere I Belong, an English-language jazz album featuring traditional Asian instrumentation. During her Ultimate Karen Mok Show tour in 2019, her performance in Lhasa, Tibet, set the Guinness World Record for the Highest Altitude Mass-Attended Music Concert. In 2024, she became the first female artist to perform a solo concert at Beijing's Bird's Nest stadium during her Big Big Show tour.

In 2024, Mok fulfilled a long-held ambition by forming the rock band Karen Mok & The Masters alongside Terry Chan, Arai, and James Lee, releasing their self-titled debut album in 2025. Beyond her performing career, Mok launched her own fragrance line, created the Mok-A-Bye Kitty multimedia character, and actively supports humanitarian and animal welfare causes through her Morris Charity Initiative and ambassadorial roles with organizations such as UNICEF. She married Johannes Natterer in Italy in 2011 and divides her time between London and her international work.
