Gyasi was born on January 2, 1980, in Hammersmith, London, to Ashanti Ghanaian parents who had moved to England in 1966. Raised alongside six siblings in Fulham and Hayes, he attended Bishopshalt School and completed his A Levels at East Berkshire College. He subsequently pursued higher education in Drama at Middlesex University.

His screen career gained momentum in 2005 when he secured his first main television role as Jeremy Hands in the ITV comedy Mike Bassett: Manager. That same year, he earned his first major film role in Shooting Dogs, directed by Michael Caton-Jones. He also expanded into stage performance, acting alongside Kit Harington in a 2008 National Theatre production of War Horse.

In 2012, Gyasi performed in the BBC Two historical miniseries White Heat as the younger version of Victor. While his performance in Cloud Atlas brought him wider attention that year, the casting choice drew criticism regarding the representation of Moriori people. He followed this with a role in the 2013 television film The Whale before starring in Interstellar. Though initially cast as the lead in BBC One's The Interceptor, a heel injury forced him to step down from the project.

Gyasi went on to land key television roles, including Major Alex "Lex" Carnahan in the 2016 CW miniseries Containment and the legendary warrior Achilles in the 2018 BBC and Netflix miniseries Troy: Fall of a City. From 2019 to 2023, he portrayed Agreus in the Amazon Prime fantasy series Carnival Row. His additional film work from this period includes leading the Western Hell on the Border, as well as roles in Cold Blood, Maleficent: Mistress of Evil, and Come Away. He also held a recurring television role as Ben Chambers in the third series of The A Word.

For his performance in Interstellar, Gyasi earned a nomination for Best Breakthrough Performance at the Black Reel Awards. Since 2023, he has starred as British Foreign Secretary Austin Dennison in the Netflix political drama The Diplomat. He resides in a Buckinghamshire village with his wife, Emma, whom he met in college and married in 1999, and they have two children.
