Loughlin began her career in the entertainment industry at the age of 11 as a print model before transitioning into professional acting during her teenage years.
At age 15, Loughlin landed her first major television role playing aspiring dancer Jody Travis on the ABC soap opera The Edge of Night, appearing on the series from 1980 to 1983. Loughlin achieved major public visibility in 1988 when she was cast as Rebecca "Aunt Becky" Donaldson Katsopolis on the ABC sitcom Full House. Initially scheduled for a six-episode arc, her character grew popular with viewers and she became a series regular, remaining with the show until its conclusion in 1995. She later reprised the character in the Netflix sequel series Fuller House from 2016 to 2018. Shortly after Full House ended, she co-starred with Tony Danza in the ABC sitcom Hudson Street from 1995 to 1996.
Throughout the 1990s and 2000s, Loughlin worked regularly across television films and series. Her credits include the television movies A Stranger in the Mirror, In the Line of Duty: Blaze of Glory, and Critical Mass, alongside guest roles on shows like Seinfeld, Spin City, and Birds of Prey.
