Beyond starring in the show, he directed more than 30 episodes and sang its closing theme song. The series ran for 11 seasons until 2004, earning critical success and numerous honors. Grammer received four Primetime Emmy Awards for Best Actor in a Comedy Series for his work on the show. He later returned to the character in a Paramount+ revival launched in 2023.

In addition to his sitcom roles, Grammer built a prominent career as a voice actor and film performer. He began voicing the recurring character Sideshow Bob on the animated series The Simpsons in 1990. His other voice projects include feature films such as Anastasia, Toy Story 2, and Teacher's Pet. On screen, he portrayed Dr. Hank McCoy, also known as Beast, in superhero films including X-Men: The Last Stand, X-Men: Days of Future Past, and The Marvels. His other film credits include Down Periscope, The Pentagon Wars, and The Expendables 3.

Grammer has maintained an active career across television drama, production, and stage performances. He won a Golden Globe Award for his dramatic portrayal of a corrupt Chicago mayor in the series Boss. On stage, he starred in Broadway revivals such as La Cage aux Folles, which earned him a Tony Award nomination, and Finding Neverland.
