Debicki was born to a Polish father and an Australian mother of Irish descent, both of whom were ballet dancers. She is the eldest of three children. She initially trained in ballet before shifting her focus to theatre after growing taller than her teachers by age twelve. She attended Huntingtower School in Melbourne, graduating as dux in 2007 with perfect study scores in English and Drama. In 2010, she earned a bachelor's degree in drama from the Victorian College of the Arts at the University of Melbourne.

Debicki made her feature film debut in the 2011 comedy A Few Best Men. Her first major screen role came as Jordan Baker in Baz Luhrmann's The Great Gatsby (2013), winning the AACTA Award for Best Supporting Actress in Film. That same year, she starred in the Sydney Theatre Company production of The Maids, winning the best newcomer award at the Sydney Theatre Awards. Her television roles followed in Rake, the Australian mystery series The Kettering Incident (2016)—for which she won an AACTA Award for Best Lead Actress in a Television Drama—and the BBC miniseries The Night Manager (2016).

Between 2015 and 2021, Debicki appeared in films including The Man from U.N.C.L.E. (2015), Everest (2015), Macbeth (2015), Breath (2017), and Guardians of the Galaxy Vol. 2 (2017), later reprising her role in Guardians of the Galaxy Vol. 3 (2023). She received critical acclaim for playing Alice in Steve McQueen's heist drama Widows (2018). Her other screen credits during this period include Vita & Virginia (2018), The Burnt Orange Heresy (2019), Christopher Nolan's spy thriller Tenet (2020), and voice roles in Peter Rabbit (2018) and Peter Rabbit 2: The Runaway (2021).

From 2022 to 2023, Debicki portrayed Diana, Princess of Wales, in the final two seasons of The Crown. Her performance in the final season earned her the Golden Globe Award, Critics' Choice Award, Screen Actors Guild Award, and Primetime Emmy Award for Outstanding Supporting Actress in a Drama Series. Her win made her the first Australian actress to earn a Primetime Emmy in that category.

Following The Crown, Debicki starred as film director Elizabeth Bender in Ti West's horror film MaXXXine (2024). In 2025, she returned to the stage in My Master Builder at Wyndham's Theatre in London. She was also cast in David Fincher's Netflix film The Further Mis-Adventures of Cliff Booth. Throughout her career, Debicki's acting approach has drawn on her dance background, incorporating physical awareness, posture, and precise movement into her performances.
