MacLaine was born in Richmond, Virginia, to Ira Owens Beaty, a psychology professor, school administrator, and real estate agent, and Kathlyn Corinne, a drama teacher from Nova Scotia. She is the older sister of filmmaker Warren Beatty. Raised in Virginia, she was enrolled in ballet classes at the Washington School of Ballet at age three to strengthen her ankles. She attended Washington-Lee High School in Arlington, Virginia, where she participated in cheerleading and theatrical productions before eventually pivoting away from ballet toward musical theater and acting.

Before graduating high school, MacLaine performed in New York City in a touring production of Oklahoma!. She made her Broadway debut in the ensemble of Me and Juliet (1953–1954) and later served as an understudy for Carol Haney in The Pajama Game. After stepping in for an injured Haney, MacLaine was spotted by Jerry Lewis and film producer Hal B. Wallis, leading to a contract with Paramount Pictures. She made her feature film debut in Alfred Hitchcock's black comedy The Trouble with Harry (1955), winning the Golden Globe Award for New Star of the Year – Actress.

MacLaine quickly rose to prominence with roles in major films such as Around the World in 80 Days (1956), Some Came Running (1958), Ask Any Girl (1959), The Children's Hour (1961), Irma la Douce (1963), and Sweet Charity (1969). Her acclaimed performance in Billy Wilder's romantic drama The Apartment (1960) earned her a Volpi Cup, a BAFTA Award, a Golden Globe Award, and an Academy Award nomination. During this era, she also became an honorary member of the Rat Pack and starred in Vittorio De Sica's episodic comedy Woman Times Seven (1967).

In the 1970s, MacLaine starred in the sitcom Shirley's World (1971–1972) and co-directed the Oscar-nominated documentary film The Other Half of the Sky: A China Memoir (1975). She returned to critical acclaim in feature films with The Turning Point (1977) and Being There (1979). A six-time Academy Award nominee for Best Actress, MacLaine won the award for her performance in the comedy-drama Terms of Endearment (1983).

Her notable later film credits include Madame Sousatzka (1988), Steel Magnolias (1989), Postcards from the Edge (1990), In Her Shoes (2005), Bernie (2011), The Secret Life of Walter Mitty (2013), Elsa & Fred (2014), and Noelle (2019). On television, she portrayed the title role in the biopic film Coco Chanel (2008) and made guest appearances in series including Downton Abbey (2012–2013), Glee (2014), and Only Murders in the Building (2022). Beyond acting, MacLaine is the author of numerous books on metaphysics, spirituality, and reincarnation, as well as the 1983 best-selling memoir Out on a Limb.

In addition to her competitive awards, MacLaine has been honored with major career achievements, including the Film Society of Lincoln Center Tribute in 1995, the Cecil B. DeMille Award in 1998, the AFI Life Achievement Award in 2012, and the Kennedy Center Honor in 2014.
