Due to her father's career, her childhood included spent time in South Carolina, Tennessee, Texas, Thailand, Germany, and France. She graduated from Marcos de Niza High School in Tempe, Arizona, and majored in drama at the University of North Carolina School of the Arts, graduating in 1986.
Parker began her acting career on the soap opera Ryan's Hope before making her film debut in Signs of Life (1989). She quickly rose to prominence in the early 1990s through notable film roles, including Grand Canyon (1991), Fried Green Tomatoes (1991), The Client (1994), Bullets Over Broadway (1994), and Boys on the Side (1995). Her later movie credits include Reckless (1995), The Portrait of a Lady (1996), The Spiderwick Chronicles (2008), Red (2010), R.I.P.D. (2013), and Red 2 (2013).
On stage, Parker made her Broadway debut in Prelude to a Kiss in 1990, earning a Tony Award nomination. She gained widespread acclaim on Broadway for her starring role in David Auburn's Proof (2001), which earned her a Tony Award for Best Actress in a Play. She achieved further Tony wins and nominations for productions such as Reckless (2005), Adam Rapp's The Sound Inside (2020), and a 2022 revival of How I Learned to Drive. In 2026, she made her London theater debut in a contemporary reimagining of The Oresteia at the Bridge Theatre.
In television, Parker earned critical recognition for portraying Harper Pitt in the HBO miniseries Angels in America (2003), winning both a Golden Globe Award and a Primetime Emmy Award. From 2005 to 2012, she starred as Nancy Botwin in the Showtime series Weeds, a performance that earned her a Golden Globe Award for Best Actress – Television Series Musical or Comedy. Her television work also includes recurring and guest roles in The West Wing, The Robber Bride, The Blacklist, and When We Rise.
