Raised in Los Angeles, California, by South Korean immigrant parents, Lee developed an interest in the performing arts while attending Harvard-Westlake School. She subsequently pursued theater studies at Northwestern University before relocating to New York City to begin her professional acting career.

Lee made her screen debut in 2006 on the television series Law & Order: Special Victims Unit. In 2007, she made her Broadway debut in the musical comedy The 25th Annual Putnam County Spelling Bee as Marcy Park. She returned to Broadway and performed on the West End in the 2010 revival of La Bête, and later appeared in the play 4000 Miles at Lincoln Center Theatre in 2011. During this period, she secured recurring television parts in High Maintenance, Nurse Jackie, and Girls, alongside roles in feature films such as Sisters (2015) and Money Monster (2016).

Her television career expanded with appearances in Wayward Pines, Chance, and The Good Fight. Lee achieved significant recognition for her portrayals of Maxine in Russian Doll and Stella Bak in The Morning Show. Her performance in The Morning Show earned her a Primetime Emmy Award nomination for Outstanding Supporting Actress in a Drama Series and two Screen Actors Guild Award nominations alongside the cast ensemble. Her television credits also include guest roles in Miracle Workers: Dark Ages, What We Do in the Shadows, and The Twilight Zone, as well as voice work in the animated series HouseBroken.

In film, Lee voiced the character Lyla in the animated feature Spider-Man: Into the Spider-Verse (2018) and its 2023 sequel Spider-Man: Across the Spider-Verse. She earned critical acclaim for her leading role as Nora in Celine Song's 2023 romantic drama Past Lives, winning the Hollywood Critics Association Midseason Film Award for Best Actress. For this role, she was also nominated for a Golden Globe Award, a Critics' Choice Movie Award, and an Independent Spirit Award.

Lee's film work further includes Problemista (2023), Tron: Ares (2025), Late Fame (2025), A House of Dynamite (2025), and voicing Lilypad in Pixar's Toy Story 5 (2026). Beyond acting, she was reported in 2021 to be developing a television adaptation of Minor Feelings for A24, and she has appeared in fashion campaigns for Loewe and Calvin Klein. Lee is married to actor and writer Russ Armstrong, with whom she has two sons.
