Hampshire was born in Montreal to her father, Robert Hampshire, who worked as a dentist. She was inspired to pursue acting at age eleven after watching a performance of Les Misérables with her mother. Her enthusiasm for the craft was reinforced by the vice principal of her all-girls Catholic school after she performed in a school play. At sixteen, she moved to Toronto to start her career in television and film. Following high school, she was accepted into the American Academy of Dramatic Arts, but she opted not to attend due to a conflicting film opportunity.
Since beginning her professional career in 1996, Hampshire has appeared in various Canadian television series, including This Space For Rent, Northern Town, and Carl². Her early film work includes portraying Margaret in Jerry Ciccoritti's feature The Life Before This, which premiered at the Toronto International Film Festival in 1999. She later played Chanel in the 2006 romantic comedy It's a Boy/Girl Thing, Alexandra in the 2009 film The Trotsky, and a starring role in the 2010 indie film Good Neighbours.
Hampshire has also worked extensively as a voice performer, contributing to animated shows such as Braceface as Alyson Malitski, Anne of Green Gables: The Animated Series as Diana Barry, Carl² as Chloe Crashman, 6teen as Starr, and Ruby Gloom as Misery. In 2014, she joined the cast of 12 Monkeys in a recurring role that was expanded to a main role for the second season. Between 2015 and 2020, she portrayed character Stevie Budd throughout the run of the CBC series Schitt's Creek.
In 2012, Hampshire starred as Jane Melman in David Cronenberg's Cosmopolis, appeared in Sean Garrity's My Awkward Sexual Adventure, and joined the cast of the futuristic zombie feature The Returned. That same year, Telefilm Canada presented Hampshire and actress Sarah Gadon with the first Birks Canadian Diamond award in Cannes. Late in 2015, she was cast in Xavier Dolan's independent drama The Death and Life of John F. Donovan.
In 2021, Hampshire starred in the series Chapelwaite and appeared in the music video for the single "Not Necessary" by The Tragically Hip. Beginning in 2022, she joined the main cast of the Amazon Prime series The Rig as Rose Mason. In 2023, she co-wrote the graphic novel Amelia Aierwood – Basic Witch, published by Z2 Comics. She was nominated for Best Voice Performance at the 14th Canadian Screen Awards in 2026 for voicing Captain Fuzzyweather in Hulu's The Bravest Knight.
