Her early film credits include John Tucker Must Die, Stay Alive, The Hitcher, The Narrows, and Chalet Girl.

Following One Tree Hill, Bush starred in the 2012 sitcom Partners and joined the main cast of Chicago P.D. in 2013, playing Detective Erin Lindsay for four seasons. She also made crossover appearances on Chicago Fire and Chicago Med. Her subsequent projects included voicing the superhero Voyd in Pixar's animated film Incredibles 2 (2018), appearing in the series Love, Victor, and serving as an executive producer and lead actor in the 2022 medical drama series Good Sam. She also hosted the podcast Work In Progress and co-hosted the One Tree Hill rewatch podcast Drama Queens.

She has led fundraising efforts following the Deepwater Horizon oil spill, supported Pencils of Promise, and participated in campaigns for Global Green, the Global Poverty Project, and I Am that Girl. A founding member of the Time's Up movement, she has advocated for LGBTQ+ rights, voting access, and reproductive freedom.
