Sharpe was born on 22 September 1986 in London to a British father and a Japanese mother. Raised in Tokyo during his early childhood, his family returned to England and settled in Surrey when he was eight years old. The oldest of three siblings, his brother Arthur Sharpe is a television and film composer. Sharpe was educated at Winchester College and studied classics at the University of Cambridge, where he served as president of the Cambridge Footlights.
After graduating in 2008, Sharpe joined the Royal Shakespeare Company for its 2008/2009 season, appearing in productions such as The Taming of the Shrew, The Merchant of Venice, and The Tragedy of Thomas Hobbes. He made his television debut playing Yuki Reid in the BBC medical drama Casualty from 2009 to 2010. During this time, he began directing and co-writing projects with Tom Kingsley, including the 2009 short film Cockroach and their 2011 debut feature film Black Pond, which earned them a nomination for the BAFTA Award for Outstanding Debut by a British Writer, Director or Producer.
In 2016, Sharpe created, wrote, directed, and starred in the Channel 4 dark comedy-drama Flowers, which ran through 2018 and received a BAFTA Television Award for best scripted comedy. He also starred in the BBC Two series Defending the Guilty from 2018 to 2019. In 2019, Sharpe portrayed Rodney Yamaguchi in the BBC drama Giri/Haji, a role that won him the BAFTA Television Award for Best Supporting Actor in 2020.
Sharpe continued directing with the 2021 biographical film The Electrical Life of Louis Wain, starring Benedict Cumberbatch, and the Sky Atlantic miniseries Landscapers. In 2022, he portrayed Ethan Spiller in the second season of the HBO series The White Lotus, earning a Primetime Emmy Award nomination for Outstanding Supporting Actor in a Drama Series. In 2023, he was named director for the film adaptation of Crying in H Mart, though he stepped down from the project by 2025.
His later credits include supporting and starring roles in the 2024 films A Real Pain and Emmanuelle, along with hosting the paranormal podcast Extrasensory. In 2025, Sharpe was cast in lead roles in the romantic comedy series Too Much and the British series Amadeus, in addition to an appearance in the upcoming series Prodigies. In his personal life, Sharpe has type two bipolar disorder. He is married to actress Sophia Di Martino, with whom he has been in a relationship since 2009, and they have two children born in 2019 and 2021.
