Clark was born in Sweden on 17 March 1989 and relocated with her family to Penarth, Wales, at two years old. Her mother is a paediatrician from North Wales, and her father is a software and security worker of Northern Irish and Glaswegian heritage. Raised alongside her younger sister, Siwan, she grew up in an English-speaking home, while her maternal grandparents spoke Welsh. She attended the Welsh-language school Ysgol Gymraeg Bro Morgannwg and became bilingual in English and Welsh.

Clark had her first acting experience playing Mrs Dai Bread in a school production of Under Milk Wood. After leaving school at sixteen while managing dyslexia and Attention Deficit Hyperactivity Disorder (ADHD), she finished her A Levels at Kings Monkton School in Cardiff. In 2009, she joined the British Youth Music Theatre production of According to Brian Haw and the National Youth Theatre of Wales before training at Drama Centre London.

Clark left Drama Centre in her final term in 2013 to star in Saunders Lewis's play Blodeuwedd with Theatr Genedlaethol Cymru. Her stage credits include Violence and Son at the Royal Court, Romeo and Juliet at the Crucible Theatre, Les Liaisons Dangereuses at the Donmar Warehouse, and King Lear at The Old Vic. In 2016, she was named one of Screen International's Stars of Tomorrow. Her early screen work included roles in the films The Call Up, Love & Friendship, Interlude in Prague, and The Man Who Invented Christmas.

In 2019, Clark appeared in the feature films Crawl, Eternal Beauty, and The Personal History of David Copperfield, and took on the title role in Saint Maud. Her performance as Maud earned her the BAFTA Cymru Award for Best Actress, alongside nominations for the British Independent Film Award for Best Actress and the BAFTA Rising Star Award. Her television roles include Sister Clara in His Dark Materials, Mina Harker in Dracula, and Bridget Conway in Murder is Easy.

Since 2022, Clark has starred as Galadriel in The Lord of the Rings: The Rings of Power, drawing on her Welsh background and bilingualism to inform her characterization. Her subsequent work includes a starring role in the folk-horror film Starve Acre and playing Ophelia in a 2025 film adaptation of Hamlet.
