He has a younger sister named Krishna. His lineage encompasses Gujarati and Turkmen background on his father's side, and Bengali and Belgian heritage on his mother's side. His maternal grandfather was Air Vice Marshal Ranjan Dutt, a Vir Chakra recipient. Shroff attended the American School of Bombay and is a dedicated devotee of Shiva, observing weekly fasts and celebrating Mahashivaratri.

With background training in martial arts, Shroff was awarded an honorary fifth-degree Taekwondo black belt in 2014 and frequently helps other performers prepare physically for film roles. Under the guidance of flexibility trainer Ziley Mawai, he prepared for his acting debut in the 2014 action romantic comedy Heropanti, produced by Sajid Nadiadwala and directed by Sabbir Khan. The film became a box-office success and earned Shroff the Screen Award for Best Male Debut as well as the IIFA Award for Star Debut of the Year – Male.

Shroff continued his screen career by reuniting with Nadiadwala and Khan for the 2016 martial arts action film Baaghi, which was commercially successful. He subsequently starred as a superhero in A Flying Jatt in 2016 and as a dancer in Munna Michael in 2017. In 2018, he headlined Baaghi 2, portraying an army officer searching for a missing child, which became one of the highest-earning Hindi releases of that year. He followed this with the 2019 college-set drama Student of the Year 2 and the high-grossing action thriller War alongside Hrithik Roshan.

In 2020, Shroff reprised his role in Baaghi 3, whose box office was affected by COVID-19 pandemic theater closures. He subsequently appeared in Heropanti 2 in 2022, played a double role in the 2023 science fiction film Ganapath, and co-starred with Akshay Kumar in the 2024 action film Bade Miyan Chote Miyan. Following these commercial disappointments, he appeared in the ensemble action film Singham Again in 2024, played the lead in the 2025 action project Baaghi 4, and was cast in the upcoming film Lag Jaa Gale.

In addition to film acting, Shroff has recorded several musical singles and joined an international performance tour in Doha in 2023. He co-founded the mixed martial arts organization Matrix Fight Night in 2019 and held an ownership and ambassadorial role with the Bengaluru Tigers in the Super Fight League. In 2024, he established Mumbay FC, joining the football club as a marquee player and competing in the Mumbai Premier League.

Shroff has represented numerous commercial brands, including Pepsi, ASICS India, Casio India, Garnier, and Prowl. His public profile led to inclusions in the Forbes India Celebrity 100 list in 2018 and 2019, Eastern Eye's dynamic decade list in 2020, and GQ's list of influential young Indians in 2024.
