Penikett was born on May 20, 1975, in Whitehorse, Yukon. He was raised in a family with three siblings; his father, Tony Penikett, served as a premier of the Yukon after relocating to Victoria, British Columbia, from England, while his mother, Lulla Sierra Johns, is an Upper Tanana woman hailing from the White River First Nation. Penikett pursued formal training in the performing arts, attending and graduating from the Victoria Motion Picture School before continuing his acting education at the Lyric School of Acting in Vancouver.

Early in his professional screen career, Penikett appeared in various Canadian television programs. He made a guest appearance on Stargate SG-1 in 2002 as a Human-Form Replicator. From 2004 to 2005, he gained regular television exposure portraying Ray Chase across eleven episodes of the police drama Cold Squad. During this period, he also featured in episodes of Smallville as two distinct characters, played a police officer in The L Word, and starred as Noah Hamilton alongside Tori Spelling in the 2005 television movie Hush.

Penikett secured a major career milestone after auditioning in Vancouver for the 2003 miniseries reimagining of Battlestar Galactica. Originally written as a minor character meant only to appear in the initial miniseries, his performance as Colonial Fleet officer Karl "Helo" Agathon impressed production leadership and test audiences alike. As a result, the character was incorporated into the full television series, becoming a recurring figure throughout its run.

Following his work on Battlestar Galactica, Penikett was cast as lead character Paul Ballard in Joss Whedon's sci-fi series Dollhouse, which debuted on the Fox network in 2009 and ran for two seasons. He also expanded his work into voice acting and interactive media, lending his voice to the antagonist Troy Hammerschmidt in the animated series Titan Maximum and portraying the rival driver Darius in the 2006 video game Need for Speed: Carbon. In 2010, he starred as Matt Ellman in the television miniseries Riverworld, based on Philip José Farmer's sci-fi books.

Penikett continued to land guest and recurring television engagements in subsequent years. He appeared as politician Jim Martin in the time-travel show Continuum and played the fallen angel Gadreel (initially introduced as Ezekiel) during the ninth season of Supernatural. His later projects include roles in the series Star-Crossed, guest starring as Michael Hastings in Criminal Minds, playing Kurtis Stryker in Mortal Kombat: Legacy, and starring in the web series Riftworld Chronicles. In 2024, he portrayed Ed Finley in the film The Killer Inside: The Ruth Finley Story.

Outside of his screen credit work, Penikett resides in Vancouver with his family. He has also engaged in charitable endeavors, notably participating in a fundraising auction alongside his Battlestar Galactica co-star Kandyse McClure to support the Babz Chula Lifeline for Artists Society.
