Cuneta was born at Our Lady of Lourdes Hospital in Santa Mesa, Manila. Her father, Pablo Cuneta, served as mayor of Pasay, while her mother, Elaine Gamboa, studied piano at the University of Santo Tomas Conservatory of Music. Raised in a musically inclined household, Cuneta learned piano, guitar, and flute as a child and frequently accompanied her aunt, performer Helen Gamboa. She made early television and film appearances before completing her education at St. Paul College, Pasig, and International School Manila.

Her commercial recording career began in 1978 with Vicor Music Corp after her uncle, producer Tito Sotto, recorded her singing "Tawag ng Pag-ibig." Later that year, she achieved wider recognition with the Rey Valera composition "Mr. DJ," earning a Gold Record and the nickname "DJ's Pet." She accumulated over a million total record sales through popular singles and multi-platinum albums such as Isn't It Romantic?, For Broken Hearts Only, and Sharon Sings Valera.

In 1981, Cuneta made her debut as a feature film lead in Dear Heart alongside Gabby Concepcion. Following its commercial success, her father partnered with executive Vic Del Rosario to establish Viva Films, releasing the follow-up project P.S. I Love You. Throughout the 1980s, Cuneta headlined major box office melodramas, including Bukas Luluhod ang Mga Tala, Dapat Ka Bang Mahalin?, and Bituing Walang Ningning, receiving multiple Box Office Queen recognitions along with honors from the FAMAS Awards and the Film Academy of the Philippines.

Cuneta also built a prominent television career, launching her musical-variety series The Sharon Cuneta Show in 1986 on IBC before its move to ABS-CBN in 1988. Through the late 1980s and 1990s, she maintained a prolific screen output in major releases such as Babangon Ako't Dudurugin Kita, Kahit Konting Pagtingin, and Maging Sino Ka Man. Her role in the 1996 Star Cinema production Madrasta earned her a grand slam of top domestic best actress awards.

During the 2000s and beyond, Cuneta continued acting in films like Magkapatid, Caregiver, and Mano Po 6, while hosting her musical talk show Sharon and launching the lifestyle magazine Sharon at Home.
