Born on November 5, 1976, in New York City, Arcelus was raised in Port Washington, New York. He is a second-generation American of Uruguayan, Italian, and Russian heritage, and he speaks Spanish fluently. His maternal grandmother was Princess Catherine Ivanovna of Russia, making him a distant relative of British Princes William and Harry. He earned his degree from Williams College.

Arcelus began his performing career in the early 2000s while spending three years working for an international business firm. During this period, he auditioned and performed in community and regional theater, including productions of The Who's Tommy, Big River, and several shows with the Weathervane Theatre repertory company, alongside a bilingual staging of West Side Story in Guatemala. He made his Broadway debut in 2002 in the musical Rent, initially serving as a swing before taking over the role of Roger in 2003. Throughout the 2000s, he also provided English-language voice dubbing for animated television programs such as Winx Club and titles in the Yu-Gi-Oh! franchise, and appeared in the 2004 movie musical Temptation.

Over the remainder of that decade and into the early 2010s, Arcelus secured prominent Broadway and off-Broadway stage roles. He originated the role of Jan in Good Vibrations in 2005, played Fiyero on tour and on Broadway in Wicked, and portrayed Bob Gaudio in Jersey Boys across multiple engagements between 2008 and 2010. He also originated the role of Buddy in the limited Broadway run of Elf the Musical, played Jack Chesney in an Encores! concert production of Where's Charley?, and originated the character of Jake Brigance in the stage adaptation of A Time to Kill at Arena Stage.

Arcelus expanded his television career significantly starting in 2012 when he was cast as Lucas Goodwin in Netflix's House of Cards, playing the recurring character across its first, second, and fourth seasons. In 2014, he joined the cast of CBS's Madam Secretary as Jay Whitman, eventually becoming a main cast member and appearing in 100 episodes through the show's conclusion in 2019. During this period, he also appeared in films like The Last Day of August, Ted 2, and Split, made guest appearances on series such as The Leftovers, FBI, Bull, and The Deuce, and co-wrote the musical Gettin' the Band Back Together as part of The Grundleshotz collective.

In late 2022, Arcelus returned to Broadway as the Baker in the revival of Into the Woods, starring alongside his wife, Stephanie J. Block, whom he married in 2007 after meeting during their time in Wicked. The couple continued their roles in the production's Broadway run into early 2023 before launching its national tour.
