He grew up in a nonreligious Jewish family with an older brother, Robert, and a younger sister, Mia. His father, Maury Apatow, worked as a real-estate developer, and his mother, Tamara Shad, worked at the music label Mainstream Records. After his parents divorced when he was 12, Apatow lived primarily with his father, and his interest in comedy developed during his childhood.

While attending Syosset High School, Apatow played jazz and created a radio program called Comedy Club on the school's station, WKWZ, which enabled him to interview prominent and emerging comedians. He began performing stand-up comedy at age 17 during his senior year. After graduating in 1985, he moved to Los Angeles to enroll in the screenwriting program at the University of Southern California, where he hosted on-campus comedy events and performed stand-up. He dropped out during his second year, shared an apartment with comedian Adam Sandler, and competed in the 1989 Johnnie Walker Comedy Search.

Apatow began his professional career writing and producing for television. In 1991, he was hired to write for the Grammy Awards, hosted by Garry Shandling, who became a key mentor. Apatow co-created and executive produced The Ben Stiller Show in 1992, earning an Emmy Award for its writing staff despite its early cancellation. He subsequently joined HBO's The Larry Sanders Show as a writer, consulting producer, and director, earning six Emmy nominations. In 1999, he executive produced the acclaimed series Freaks and Geeks, followed by creating and executive producing Undeclared in 2001.

His major breakthrough in feature films occurred in the mid-2000s. Apatow produced the successful comedy Anchorman: The Legend of Ron Burgundy in 2004 before making his feature directorial debut with The 40-Year-Old Virgin in 2005, which he also co-wrote and produced. In 2007, he wrote, directed, and produced Knocked Up, which achieved major commercial and critical success. During this period, he also produced several comedy hits through his company, Apatow Productions, including Superbad, Pineapple Express, Forgetting Sarah Marshall, and Step Brothers, while co-writing Walk Hard and You Don't Mess with the Zohan.

Apatow continued his filmmaking career by writing, directing, and producing Funny People in 2009, followed by This Is 40 in 2012, Trainwreck in 2015, The King of Staten Island in 2020, and The Bubble in 2022. As a producer, his film credits expanded to include Get Him to the Greek, Bridesmaids, Begin Again, and The Big Sick. On television, he developed and produced series such as Funny or Die Presents, Girls, Love, and Crashing.

Throughout his career, Apatow has earned significant recognition for his work as a writer, director, and producer. He has been nominated for eleven Primetime Emmy Awards, winning three, and five Writers Guild of America Awards, winning one. His accolades also include nominations for two Producers Guild of America Awards, a Golden Globe Award, and a Grammy Award.
