Comer was born on 11 March 1993 in Liverpool, where she was raised in the Childwall suburb. She attended St Julie's Catholic High School, where she became close friends with future Olympic athlete Katarina Johnson-Thompson. At age 11, Comer began acting at CALS Theatre School. In 2006, she won her category at the Liverpool Performing Arts Festival with a monologue about the Hillsborough disaster. After performing the monologue at her school talent show, her drama teacher arranged an audition for a BBC Radio 4 play, marking her first professional acting job.
Her professional screen career began in 2008 with a guest role on The Royal Today, followed by her stage debut in The Price of Everything in 2010. She subsequently appeared in television series such as Justice, Silent Witness, Remember Me, and My Mad Fat Diary. Comer achieved further recognition in Doctor Foster and starred in the miniseries Thirteen, receiving a BAFTA Television Award nomination. She also appeared in Rillington Place, The White Princess, and made her feature film debut in England Is Mine in 2017.
Comer gained widespread global recognition playing sociopathic assassin Villanelle in the BBC America spy thriller series Killing Eve from 2018 to 2022. For her performance, she won a BAFTA Television Award and a Primetime Emmy Award in 2019. In film, she starred as Millie and Molotov Girl in Free Guy, and as Marguerite de Carrouges in Ridley Scott's historical drama The Last Duel. For her performance as care home worker Sarah in the television drama Help, which she executive produced, Comer earned her second BAFTA Television Award.
In 2022, Comer made her West End debut in Suzie Miller's one-woman play Prima Facie as defense barrister Tessa Ensler. She transferred with the production to Broadway in 2023. Her performance won both the Laurence Olivier Award for Best Actress and the Tony Award for Best Actress in a Play. A filmed performance of the play was released in cinemas worldwide by National Theatre Live.
Comer's later film projects include Jeff Nichols's drama The Bikeriders, the environmental thriller The End We Start From—which she also executive produced—and Danny Boyle's horror film 28 Years Later. She also voiced Emily Hartwood in the 2024 video game Alone in the Dark. Off-screen, she was named the face of the Loewe spring/summer 2020 campaign and was included in Forbes' 30 Under 30 Europe list in 2019.
