Pannu was born on August 1, 1987, in New Delhi to a Jat Sikh family. Her father worked as a real estate agent prior to his retirement, and her mother is a homemaker. Growing up alongside a younger sister, her family survived the 1984 anti-Sikh riots in Delhi after finding safety with neighboring families. She completed her schooling at Mata Jai Kaur Public School in Ashok Vihar and later earned a degree in Computer Science and Engineering from Guru Gobind Singh Indraprastha University.

After completing her university studies, Pannu worked as a software engineer before embarking on a modeling career following her selection in a television talent competition in 2008. She appeared in numerous print and broadcast commercials, endorsing prominent commercial brands and winning contest titles. Seeking broader professional recognition, she transitioned into acting, making her feature debut in the 2010 Telugu musical drama Jhummandi Naadam. She made her Tamil film debut in Aadukalam (2011) and entered Hindi cinema through the comedy Chashme Baddoor (2013).

Following appearances in several South Indian projects, Pannu achieved wider recognition in Hindi cinema for her performance in the spy thriller Baby (2015) and its spin-off Naam Shabana (2017). Her career marked a major breakthrough with the critically acclaimed courtroom drama Pink (2016). She subsequently expanded her body of work with leading roles in the naval war drama The Ghazi Attack, the sports film Soorma, the legal drama Mulk, and the romantic film Manmarziyaan.

Pannu continued to achieve critical and financial success through various high-profile releases. She starred in the psychological mystery Badla, the home-invasion thriller Game Over, and the space drama Mission Mangal. Her portrayal of sharpshooter Prakashi Tomar in the biographical drama Saand Ki Aankh won her the Filmfare Critics Award for Best Actress. She later received the Filmfare Award for Best Actress for her performance as a woman seeking a divorce in the social drama Thappad.

In her subsequent career, Pannu featured in several digital streaming productions, including the romantic thriller Haseen Dillruba, its sequel Phir Aayi Hasseen Dillruba, Rashmi Rocket, and Looop Lapeta. She also starred in the comedy-drama Dunki (2023), which became the highest-grossing release of her career.

Outside of feature films, Pannu engages in commercial ventures and sports ownership. She is a co-owner of the event management enterprise The Wedding Factory as well as the badminton team Pune 7 Aces, which competes in the Premier Badminton League. She is married to former professional badminton player Mathias Boe.
