Raised in an evangelical Christian household, Lopilato grew up with her older siblings, actor Darío and actress-nutritionist Daniela. Her parents are Eduardo Lopilato and Beatriz de la Torre, and her background includes Spanish and Italian heritage. She began appearing in television commercials at five years old and took up child modeling at seven. Following small appearances on television shows such as Mi familia es un dibujo, she landed her breakthrough role in Chiquititas at age twelve, subsequently recording multiple soundtrack albums, starring in a miniseries spin-off, and appearing in the 2001 film Chiquititas: Rincón de Luz.

The cast members formed the musical group Erreway, releasing studio albums including Señales, Tiempo, and Memoria, which achieved certified gold and platinum status in Argentina and sold over five million copies worldwide. After starring in the group's 2004 movie Erreway: 4 Caminos, she eventually departed the band and signed with Universal Music Group for solo work. She later starred in the series Atracción x4 with her brother Darío, generating another musical ensemble that earned gold certifications.

Lopilato transitioned into adult comedy and drama roles, prominently playing Paola Argento in the Argentine adaptation Casados con Hijos, which earned her a nomination for a Martín Fierro Award. She expanded her career through projects such as Alma Pirata, El Capo, Encandilados, and a stage role in Arlequín that earned an ACE Award nomination. In addition to co-hosting the Latin MTV Awards, she starred in feature films such as Papá por un Día, the animated production Plumíferos, and the 2011 Spanish film Predeterminados, marking her international screen debut.

Her subsequent acting work included television projects like Lobo, the Italian miniseries A Good Season, and the Argentine drama En terapia, which brought her a second Martín Fierro Award nomination. She went on to feature in films such as Las Insoladas, Los que aman, odian, and her first North American production, Fair Market Value.
