Raised in New Orleans, Louisiana, Thornton grew up with an amateur opera singer father who performed in local theater. While studying as a theater major at the University of New Orleans, Thornton acted in a variety of stage plays. After two years of college, he chose to move to Los Angeles to further his artistic training at the Stella Adler Academy.

Thornton made his feature film debut in 1990 with a role in the horror movie *Watchers II*. In 1992, at the age of 25, he experienced a severe rock climbing accident that fractured two vertebrae, resulting in paralysis from the waist down. Following his injury, he focused primarily on stage acting throughout the rest of the decade.

In 2000, Thornton earned widespread praise for his performance as the title character in a Lillian Theater production of *Hamlet* in Los Angeles. His portrayal was noted as likely being the first time the famous Shakespearean protagonist was performed by an actor using a wheelchair.

Thornton wrote and starred in the 2010 feature film *Sympathy for Delicious*, portraying a disc jockey named Dean O'Dwyer. The movie was directed by his close friend Mark Ruffalo, who also co-starred alongside Orlando Bloom, Laura Linney, and Juliette Lewis.

Over the course of his television career, Thornton secured recurring parts in series such as *Any Day Now*, *Rules of Engagement*, and *Vice Principals*. He also made guest appearances on prominent shows including *Curb Your Enthusiasm*, *Alias*, *Brothers & Sisters*, *My Name Is Earl*, *Grey's Anatomy*, and *Will & Grace*.

From 2018 until 2024, Thornton played his best-known recurring role as Kenny "Shammy" Shamberg, a paralyzed Marine veteran who served in Iraq, on the CBS reboot of *Magnum P.I.* Looking ahead, he has been cast in the upcoming Netflix limited series adaptation of S. A. Cosby's novel *All the Sinners Bleed*.
