Greene was born in London to Welsh DIY expert Harry Greene and English actress Marjie Lawrence. Raised alongside her younger sister, presenter Laura Greene, she was educated at Gospel Oak Primary School and the Grey Coat Hospital School in London while pursuing child acting opportunities. She went on to graduate with a degree in Drama from the University of Hull before seeking acting roles in Birmingham, Manchester, and London.
In January 1980, Greene appeared in the daytime drama Together. After performing in a BBC adaptation of Pamela Brown's The Swish of the Curtain, she was chosen by editor Biddy Baxter to succeed Tina Heath as a presenter on Blue Peter. At age 22, Greene became the show's youngest presenter at the time, filming in Cornwall within two weeks of joining. She co-presented the series from May 1980 to June 1983, a period during which the show won a BAFTA Award.
Following her departure from Blue Peter, Greene presented Saturday Superstore alongside Mike Read and subsequently co-hosted Going Live! with Phillip Schofield. Her work on Going Live! earned her the SOS Award for Best Female on TV for three consecutive years. During this period, she appeared in television commercials for Nescafé and presented Channel 4's daytime antiques show Collectors' Lot. In September 1988, Greene and her partner, presenter Mike Smith, survived a serious helicopter accident in Gloucestershire, returning to host Going Live! while still recovering in plaster.
Alongside her primary presenting career, Greene acted in television series including the Doctor Who serial Attack of the Cybermen, Casualty, Brookside, and French & Saunders. She co-hosted the live series Hospital Watch in 1991, the 1992 hoax paranormal broadcast Ghostwatch, and the 1993 game show Happy Families. In 1997, Greene and Smith appeared in the feature film The Man Who Knew Too Little starring Bill Murray. Beginning in 1996, she hosted a daily news and debate series for the Carlton Food Network for four years.
Greene continued her television career with projects such as Discovery Home & Health's Weddings Live from 2005 to 2007 and competed on Dancing on Ice in 2008. She traced her family ancestry back to 17th-century Wales in a 2010 episode of Coming Home. Following the death of her husband Mike Smith in 2014, Greene guest-presented editions of This Morning in 2017. In 2023, she began co-hosting the BBC One quiz show The Finish Line alongside Roman Kemp. She has also served as an ambassador for Target Ovarian Cancer since 2012.
