His career in the entertainment industry includes early appearances in live-action television projects. He took on a voice role in Toshiie to Matsu and made his debut in a television series with Fafner in the Azure: Dead Aggressor in 2004. He also performed as Muromachi Toji in the theatrical production of The Prince of Tennis musical.

Over the course of his performing career, Ono has voiced numerous primary characters across various popular anime series. Some of his prominent roles include Tetsuya Kuroko in Kuroko's Basketball, Giorno Giovanna in JoJo's Bizarre Adventure, Yuya Sakaki in Yu-Gi-Oh! Arc-V, and Riku Nanase in Idolish7. Additional television anime credits feature roles such as Ryuunosuke Akutagawa in Bungo Stray Dogs, Slaine Troyard in Aldnoah.Zero, Hakuryu Ren in Magi: The Labyrinth of Magic, and Mikaela Hyakuya in Seraph of the End.

In addition to original Japanese animation, Ono is recognized for providing the voice of major imported film characters for Japanese audiences. He served as the Japanese dub actor for the title character throughout the Harry Potter motion picture franchise. He also voiced the character Miles Morales in the animated films Spider-Man: Into the Spider-Verse and Spider-Man: Across the Spider-Verse.

Outside of acting, Ono initiated a music career in February 2014 by issuing his debut single titled Fantastic Tune. His musical catalog includes four singles, two mini-albums, and one full-length studio album. Under the Lantis label, he released a fifth single called Five Star on June 27, 2018, which was followed by a live summer performance event named Ono Kensho Live Tour 2018.

Throughout his professional path, Ono was previously affiliated with Sunaoka Office, an entity connected to Gekidan Himawari, before joining the talent management agency Animo Produce. His contributions to the voice acting industry were formally honored in 2022 when he earned the Best Lead Actor Award during the 16th Seiyu Awards.

In his personal life, Ono's relationship with fellow performer Kana Hanazawa was publicly confirmed in February 2017 during a live streaming event following media reports. The couple announced their marriage on July 8, 2020. Later, on September 14, 2025, both individuals shared statements on their respective social media accounts announcing their divorce, attributing the separation to divergent lifestyles and a lack of available time together.
