A significant portion of her career features creative collaborations with her husband, director and writer Mike Flanagan.

Siegel was raised in Silver Spring, Maryland, and is of Russian-Jewish, Polish-Jewish, and German-Jewish lineage. She completed her higher education at Syracuse University, graduating in 2004. In her personal life, Siegel publicly shared in 2008 that she is bisexual. She married Mike Flanagan in early 2016. The couple has two children, a son named Cody and a daughter named Theodora, and Siegel is also stepmother to Flanagan's son from a previous relationship.

Siegel made her screen debut in the early 2007 release The Curse of The Black Dahlia. Later that year, she starred in Hacia La Oscuridad, which premiered at the Tribeca Film Festival, and appeared alongside Ruby Dee and Chelsea Handler in Steam. She followed these with a role in the 2008 short film Knocked Down and television appearances on Ghost Whisperer, Numb3rs, Castle, and Mob City. Her early feature credits also include the drama-thriller Wedding Day, the comedy Man Camp, and the horror film Demon Legacy.

Her ongoing work in horror deepened with the 2013 film Oculus, directed by Flanagan. In 2016, Siegel starred as a deaf-mute protagonist in the home-invasion thriller Hush, a project that marked her feature screenwriting debut alongside Flanagan. That same year, she starred in Flanagan's Ouija: Origin of Evil and appeared in a television commercial for Stelara. In 2017, she appeared in the Netflix film adaptation of Stephen King's Gerald's Game.

In television, Siegel achieved further prominent roles in several Netflix horror series, beginning with her portrayal of Theodora Crain in The Haunting of Hill House in 2018. She subsequently starred in The Haunting of Bly Manor in 2020, played Erin Greene in Midnight Mass in 2021, and appeared in The Fall of the House of Usher in 2023.

Beyond screen acting, Siegel narrated the podcast Calling Darkness and served as a voice actor for The NoSleep Podcast. She also collaborated on plans to adapt the young adult novel 13 Days to Midnight. In 2024, she starred in the film The Life of Chuck and made her debut as a director by contributing a segment to the horror anthology film V/H/S/Beyond.
