On television, she is widely known for her role as Adèle in the acclaimed series Les Revenants.

Hesme was born on July 30, 1979, in Troyes, located in the interior region of Aube, France, to parents who worked as civil servants. Raised in a creative family, her sisters Annelise Hesme and Élodie Hesme also pursued careers in acting. She formally trained in acting at the Conservatoire National Supérieur d'Art Dramatique in Paris, performing in numerous theatrical productions during her studies.

Her screen debut occurred in the 1999 short feature Dieu, que la nature est bien faite!. While performing in a Parisian stage play, she was spotted by director Jérôme Bonnel, who subsequently cast her in the 2002 movie Le Chignon d'Olga. In the early stages of her screen career, she played supporting parts in feature films such as Focus and À ce soir while continuing her work on the French stage.

Hesme secured her first major cinematic lead role starring alongside Louis Garrel in Philippe Garrel's 2005 film Regular Lovers. She followed this project with the 2006 short production Comment on freine dans une descente?. In 2007, she reunited with Louis Garrel for Christophe Honoré's Love Songs. She later appeared in Honoré's 2009 work The Beautiful Person and joined an international cast for Raúl Ruiz's 2010 Portuguese production Mysteries of Lisbon.

In 2011, Hesme portrayed the titular character in the feature Angèle et Tony. She followed this in 2012 by starring in Catherine Corsini's film Three Worlds, which competed within the Un Certain Regard segment at the Cannes Film Festival. That same year, she joined the main cast of the television drama Les Revenants, a series that achieved broad international acclaim from audiences and reviewers.

Throughout her career, Hesme has received several industry honors. Her performance in Love Songs earned her the SACD Award for Female Revelation in 2008, alongside a nomination for the César Award for Most Promising Actress. She was also nominated for a Molière Award in 2008 for her stage performance in Marivaux's La Seconde Surprise de l'amour. In 2011, she won the César Award for Most Promising Actress for her lead role in Angèle et Tony.
