Throughout his career, he collaborated with filmmakers such as Antonio Margheriti, Marino Girolami, Paolo Solvay, Alfonso Brescia, and Godfrey Ho, appearing alongside actors including Klaus Kinski, Anita Ekberg, Helmut Berger, and Michèle Mercier.

Harrison was born in Salt Lake City, Utah, and relocated to Los Angeles at age 17. He worked at the Vic Tanny and Bert Goodrich gyms, where interactions with film industry figures who trained there inspired him to study acting. In 1961, he married Loretta Nicholson, the daughter of American International Pictures co-chief James H. Nicholson. Harrison began his acting career in Santa Monica stage productions, television, and minor feature film parts. While studying under acting coach Sandy Meisner at Twentieth Century Fox, he made an appearance in South Pacific.

After signing a three-picture deal with American International Pictures, Harrison relocated to Italy in the early 1960s. His formal acting training set him apart from other muscular American actors in Italy at the time. He made his Italian film debut and landed his first starring role in The Invincible Gladiator (1962). Harrison became a prominent lead in sword-and-sandal films, Eurospy features such as Secret Agent Fireball (1965) and its 1966 sequel Killers Are Challenged, and Spaghetti Westerns like Gunfight at Red Sands (1963). Notably, he turned down the lead in A Fistful of Dollars and recommended Clint Eastwood for the role.

As Spaghetti Westerns declined in the 1970s, Harrison transitioned to low-budget international productions filmed in Egypt, Turkey, Yugoslavia, and Hong Kong with the Shaw Brothers. He also directed several films in the early 1970s, including the comedic Spaghetti Western Two Brothers in Trinity (1972), in which he co-starred with Donald O'Brien. His other notable projects from this decade included Churchill's Leopards (1970), Acquasanta Joe (1971), Dig Your Grave, Friend...Sabata's Coming (1971), and the crime thriller La Belva Col Mitra (1977).

During the 1980s, Harrison starred in low-budget action films produced in the Philippines for Silver Star Film Company, where he improvised scenes and wrote screenplays under pseudonyms. In Hong Kong, he worked with filmmaker Godfrey Ho, who re-edited Harrison's scenes into at least 24 different martial arts ninja films, including Ninja Terminator and Cobra Vs. Ninja. Although Harrison was dissatisfied with this outcome, these releases developed a cult following. Disgusted by the process, he returned to the United States.

In the 1980s and 1990s, Harrison took on diverse roles, portraying American President Andrew Johnson in the Iranian production Hajji Washington (1982) and appearing in films like Amok (1982), Dark Mission (1987), and Lies of the Twins (1991). His final screen appearances were in Angel Eyes (1993) and Jerks (2000). Outside of acting, he founded the electronics company Gladiator Electronics with his son Sebastian. In January 2025, his Malibu home was destroyed in the Palisades Fire.
