Lyles was born on July 18, 1997, in Gainesville, Florida, to Keisha Caine Bishop and Kevin Lyles, both of whom competed in track and field at Seton Hall University. He grew up with two siblings, Josephus and Abby, and relocated to Alexandria, Virginia, following his parents' divorce. Lyles was homeschooled during his early years while dealing with asthma before attending T. C. Williams High School (now Alexandria City High School). Originally a gymnast, he took up track and field at age 12.

In 2014, Lyles won gold in the 200 m at the Youth Olympic Games. He was named the 2015 high school boys athlete of the year by Track & Field News after clearing 2.03 m in the high jump. Later that year, he won gold in both the 100 m and 4 × 100 m relay at the World U20 Championships.

Lyles turned professional in July 2016, signing with Adidas alongside his brother Josephus and foregoing a college commitment to the University of Florida. In 2017, he won the 300 m title at the USA Indoor Championships in an indoor world record time of 31.87 seconds, and secured a silver medal in the 4 × 200 m relay at the World Relays. In 2018, Lyles became the youngest U.S. 100 m champion since 1984 and recorded four sub-19.70 performances in the 200 m.

Lyles earned his first global senior titles at the 2019 World Athletics Championships in Doha, winning gold in the 200 m and 4 × 100 m relay. At the 2022 World Championships in Eugene, he broke Michael Johnson's long-standing American record in the 200 m with a time of 19.31 seconds to win gold, while also earning a silver in the 4 × 100 m relay. At the 2023 World Championships in Budapest, Lyles completed a sprint triple by winning gold in the 100 m, 200 m, and 4 × 100 m relay. At the 2024 Olympic Games, he won the 100 m gold medal with a personal best of 9.784 seconds and added a bronze medal in the 200 m.

At the 2025 World Championships in Tokyo, Lyles won bronze in the 100 m, captured his fourth consecutive 200 m title, and anchored the winning U.S. 4 × 100 m relay team. In June 2026, he set a 150 m curved track world best of 14.67 seconds at the Ostrava Golden Spike.
