She is widely recognized for her television and radio work, including regular panel show appearances and her long-running role as the presenter of The Great British Bake Off: An Extra Slice. In 2003, she was listed in The Observer as one of the 50 funniest acts in British comedy.

Brand was born in Clapham, London, and grew up in Hastings, East Sussex, as well as the Kent villages of St Mary's Platt and Benenden. Her mother worked as a social worker and her father was a structural engineer. After attending schools including Tunbridge Wells Girls' Grammar School, Hastings High School for Girls, and Bexhill College, she held jobs working in a pub, for Barnardo's, and as a nursing assistant. She earned a joint social science degree with a Registered Mental Nurse qualification at Brunel University and worked as a psychiatric nurse for ten years at South London Bethlem hospital, Cefn Coed Hospital, and Maudsley Hospital.

Beginning her comedy career under the stage name the Sea Monster, Brand performed in London alternative comedy clubs and gained early television exposure on Saturday Live. Her performance style involved delivering jokes in a bored monotone with pauses in between, drawing heavily on popular culture and public figures. In 1993, she joined BBC's The Brain Drain as a resident panellist, followed by her own Channel 4 series, Jo Brand Through the Cakehole, co-written with Jim Miller. She also made frequent appearances on BBC Radio 4 programmes such as The News Quiz and Just a Minute.

Over her career, Brand became a staple of British television. She has made guest appearances on panel shows such as Have I Got News for You, Would I Lie to You?, and QI, where she became the most frequent guest with 34 appearances. She has also appeared on Countdown as a Dictionary Corner guest 88 times. Brand co-created, co-wrote, and starred in the NHS satire sitcom Getting On opposite Joanna Scanlan and Vicki Pepperdine. Her other screen projects include writing and starring in the comedy series Damned, writing a feature-film adaptation of her novel The More You Ignore Me, and presenting Jo Brand's Commercial Breakdown, Jo Brand's Big Splash, and Jo Brand's Cats & Kittens.

Beyond broadcasting, Brand is active in charitable causes. She serves as an ambassador for the Alzheimer's Society, president of the Ectopic Pregnancy Trust, and patron of several organisations including the National Self Harm Network and International Animal Rescue. In 2016, she completed a 150-mile walk across Britain for Sport Relief, which was documented in the special Jo Brand's Hell of a Walk for Sport Relief.

Brand's contributions have earned her prominent industry recognitions. She won the 2011 BAFTA award for Best TV Comedy Actress for her role in Getting On. In addition to a Journal of Psychiatric and Mental Health Nursing Lifetime Achievement Award in 2007, she has received honorary doctorates and degrees from the University of Glamorgan, Canterbury Christ Church University, and the University of Suffolk.
