Williamson was born in Milton Keynes and grew up in Newport Pagnell, where she attended Portfields School for primary education and Ousedale School for secondary education. Coming from a family divided in football support, Williamson supported Arsenal alongside her mother and grandmother. She began her youth career with Scots Youth FC as the only girl on the team before joining the Rushden & Diamonds Centre of Excellence at age six. In 2006, at age nine, she transitioned to Arsenal's youth programme after following her coach to the club.

Williamson made her senior team debut for Arsenal in March 2014 as a teenager during a Women's Champions League quarter-final match against Birmingham City. She quickly established her presence, winning her first major title with Arsenal in the 2014 FA Women's Cup final and scoring her first professional goals later that year. Initially playing in midfield, Williamson shifted to central defence under manager Joe Montemurro in late 2017 to align with Arsenal's tactics of playing out from the back. She played a key role in Arsenal's 2018–19 league title win, leading the squad in touches, completed passes in the opposition half, and interceptions. After making her 150th club appearance in 2020 and reaching 200 appearances in December 2022, Williamson agreed to multiple contract extensions, eventually breaking the record for total WSL appearances for Arsenal.

Over the course of her tenure at Arsenal, Williamson has won the Women's Super League title once, the UEFA Women's Champions League once, two FA Cups, and four League Cups. Despite physical setbacks, including ankle surgery in 2016, a ruptured ACL in 2023, and knee surgery in 2025, she returned to play each time. Her club accomplishments included winning the 2025 Champions League following a 1–0 victory over Barcelona.

At the international level, Williamson represented England across youth teams starting in 2010, captaining the under-17 side at the 2013 UEFA Championship. She made her senior debut in 2018 during 2019 FIFA World Cup qualifying and represented Great Britain at the 2021 Summer Olympics. After becoming a regular starter under manager Sarina Wiegman, Williamson was appointed permanent England captain in 2022. She led England to victory at UEFA Euro 2022, earning selection to the Team of the Tournament, and captained the squad again for their Euro 2025 title, becoming the first England captain to lift two major trophies.
