McKeon was born in Wollongong, New South Wales, Australia, into a prominent swimming family. Her father, Ron McKeon, is a two-time Olympian and four-time Commonwealth gold medalist, while her mother Susie and uncle Rob Woodhouse also competed at elite levels. Her brother, David McKeon, is an Olympian as well. McKeon completed her secondary education at The Illawarra Grammar School in 2012. In 2014, she relocated to Brisbane to train under coach Vince Raleigh at the Chandler Aquatic Centre, later moving to train under Michael Bohl in 2015. She subsequently pursued a bachelor's degree in public health and health promotion, majoring in nutrition, at Griffith University on the Gold Coast.
McKeon began competing internationally at the 2010 Summer Youth Olympics in Singapore, where she earned six medals, including gold in the girls' 4 × 100 metre medley relay. After narrowly missing selection for the 2012 London Olympic Games by finishing seventh in the 100 metre freestyle at the Australian trials, she briefly took a break from the sport to rejuvenate her drive before returning to elite competition.
She made her Olympic debut at the 2016 Rio de Janeiro Games alongside her brother David, making them the first brother and sister to swim for Australia at an Olympic Games since 1960. In Rio, McKeon secured a gold medal in the 4 × 100 metre freestyle relay in world record time, along with two silvers and a bronze in the 200 metre freestyle. At the delayed 2020 Tokyo Olympics, she became the most decorated athlete across all sports at the Games, winning four gold and three bronze medals. Her Tokyo performance included Olympic records and gold medals in both the 50 metre and 100 metre freestyle events.
In addition to her Olympic success, McKeon built an extensive record at the World Aquatics Championships, capturing 20 medals—including five golds—across the 2013, 2015, 2017, 2019, and 2023 editions. At the Commonwealth Games in 2014 and 2018, she tied and set competition records by securing a total of 20 medals, including 14 golds. She was also the overall highest-scoring competitor at the 2021 FINA Swimming World Cup, collecting 14 medals.
At the 2024 Summer Olympics in Paris, McKeon earned three additional medals: gold in the 4 × 100 metre freestyle relay, silver in the 4 × 100 metre medley relay, and bronze in the mixed 4 × 100 metre medley relay. The gold medal marked her sixth career Olympic gold, setting the all-time Australian record. Following her final swim in Paris, McKeon officially confirmed her retirement from competitive swimming on 25 November 2024.
