Oreiro was born on May 19, 1977, to Carlos Florencio Oreiro Poggio and Mabel Cristina Iglesias Bourié. Of Spanish and Italian heritage, she grew up alongside an older sister. After spending two years in Málaga, Spain, during her childhood, her family returned to Uruguay, where she attended elementary schools in Montevideo's Villa del Cerro neighborhood. She began studying drama at age eight and started appearing in television advertisements by age twelve, eventually appearing in over thirty commercial campaigns.

At age sixteen, Oreiro relocated to Buenos Aires, Argentina, to pursue her artistic career. In 1993, she won a children's competition on El Show de Xuxa, becoming an entertainer and assistant on the program. She subsequently worked as an MTV VJ and secured early roles in Argentine television productions, including Dulce Ana, 90-60-90 modelos, and her stage debut in Butterflies Are Free. She landed her first leading telenovela role in 1997's Ricos y Famosos, followed by her feature film debut in An Argentinian in New York in 1998.

Following her cinematic debut, Oreiro launched her self-titled debut album, featuring the single "Cambio Dolor," which served as the theme for Muñeca Brava. The telenovela's global popularity brought her massive fan bases in Russia and Israel, where she earned acting and music honors. She expanded her discography with the albums Tu Veneno and Turmalina, scoring a Latin Grammy nomination for Best Female Pop Vocal Album and being crowned Queen at Chile's Viña del Mar Festival in 2000. Over her musical career, she has sold over ten million records globally.

Transitioning toward feature films in the late 2000s, Oreiro solidified her reputation as a leading dramatic and comedic actress. She earned critical acclaim for her roles in films such as Music on Hold, Miss Tacuarembó, My First Wedding, and Super Crazy. Her performances in the historical dramas Clandestine Childhood and The German Doctor garnered widespread acclaim, with both films being shortlisted for the Academy Awards. She also portrayed the biographical lead in I'm Gilda, recorded its soundtrack album, and starred in titles such as Expiration Date and The Woman in Line.

In addition to her film and music work, Oreiro returned to television with celebrated roles in prime-time series like Sos mi vida, Solamente vos, and Entre caníbales. She expanded her career into television presenting, hosting major talent shows such as Got Talent Uruguay, La Voz Uruguay, and ¿Quién es la Máscara? Argentina.
