Reclassifying in 2015, he joined Kentucky under coach John Calipari. In his freshman season, Murray averaged 20.0 points per game—a program record for freshmen—earning third-team All-American, All-SEC First Team, and SEC All-Freshman Team honors.
Selected seventh overall in the 2016 NBA draft by the Denver Nuggets, Murray signed his rookie contract in August 2016. He was named Western Conference Rookie of the Month for October and November, earned MVP honors at the 2017 Rising Stars Challenge, and was named to the NBA All-Rookie Second Team. Over his next two seasons, he established himself as a primary scorer for Denver, recording a career-high 48 points against the Boston Celtics in November 2018 and making his playoff debut in 2019.
After signing a five-year contract extension in 2019, Murray starred during the 2020 postseason in the NBA bubble. He posted two 50-point playoff games against the Utah Jazz and a 40-point Game 7 performance against the Los Angeles Clippers to reach the Western Conference Finals. In February 2021, Murray scored 50 points against Cleveland without a free throw attempt, an NBA first. However, he suffered a torn ACL in April 2021 that required surgery and sidelined him for the entire 2021–22 season.
Murray returned to the floor in October 2022 and became the Nuggets' all-time leader in three-pointers made during the 2022–23 regular season. In the 2023 playoffs, he helped lead Denver to its first NBA championship, highlighted by averaging over 20 points and 10 assists in the Finals against the Miami Heat. In the 2024 postseason, Murray hit two game-winning shots in a single first-round series against the Los Angeles Lakers.
