She was raised as the daughter of Fernando Zegers Hochschild, a notable practitioner in gynecology, and Mónica Oportot Salbach, an adventurous photographer and Buddhist. She grew up alongside a younger sister named Emilia. Zegers received her early schooling at Saint George's school in Santiago before pursuing professional drama training at the Gustavo Meza Theater School, an institution later known as Teatro Imagen.

Her acting career on screen commenced in 1995 with her film debut in Christine Lucas's feature En tu casa a las ocho. Over the following decades, she built a cinematic body of work comprising roughly a dozen feature films. She frequently collaborated with prominent Chilean directors, appearing in nearly all projects helmed by Pablo Larraín, as well as productions directed by Matías Bize and Marialy Rivas.

Zegers gained wide public recognition on television through her extensive appearances in soap operas broadcast by Chilean National Television. Her television work encompasses titles such as Iorana, La Fiera, Romané, Pampa Illusion, El circo de las Montini, Puertas adentro, Los Pincheira, and Amor por accidente. Among her various screen roles, she has cited her portrayal of María Jacobé in Romané and her performance as Asunción in Los Pincheira as her personal favorites.

Throughout her movie career, Zegers has featured in a succession of highly honored Chilean films. Her motion picture credits include Tony Manero in 2008, Post mortem in 2010, La vida de los peces in 2010, and No in 2012. She subsequently appeared in El club in 2015, the 2017 film A Fantastic Woman—which secured the Academy Award for Best Foreign Language Film—and the 2023 movie El Conde. For her dramatic work in the feature Los Perros, she received individual honor, earning the Havana Star Prize for Best Actress at the 19th Havana Film Festival New York.

In addition to her film and television work, Zegers has performed in a variety of stage productions. Her theater credits include roles in plays such as Provincia Kapital, Pecados, Madre, Numancia, and Fin del eclipse.

Regarding her personal life, Zegers was in a relationship with actor Ricardo Fernández between 2001 and 2004. In 2006, she met film director Pablo Larraín, and the two married in 2008. The couple had two children together prior to their separation in late 2014.
